Rat cocaine- and amphetamine-regulated transcript (CART) is a 117-aa anorexigenic neuropeptide cleaved from pro-CART in hypothalamic, amygdalar and striatal neurons. It targets pre-synaptic voltage-gated Ca²⁺ channels and G-protein-coupled receptors, dampening dopamine release while activating anorexigenic melanocortin pathways. Its N-terminal disulfide-bonded loop presents a unique, species-restricted epitope ideal for antibody generation; anti-CART immunoreagents detect intact peptide and major processed fragments in brain lysates, CSF and cultured neurons without cross-reacting with related peptides, providing a precise read-out of reward and appetite circuits.
