Rat chondroitin sulfate proteoglycan 5 (CSPG5, NG2) is a 232 kDa single-pass transmembrane protein whose central 1,500-residue domain carries ~30 covalently linked chondroitin sulfate chains that create a dense anionic coat around oligodendrocyte progenitors and pericytes. A solvent-exposed 20-aa "QEK" loop (aa 1,410-1,429) between CS attachment sites folds into a β-hairpin unique among rat CSPGs, presenting a conformational epitope preserved after chondroitinase and fixation. Affinity-purified antibodies targeting this loop trace native CSPG5 in cerebral white matter, vascular mural cells, and sciatic nerve, delivering crisp membrane trails and single-band clarity on immunoblots with no cross-reaction to CSPG4 or neurocan.
