Target Information
Rat bone morphogenetic protein 3 (BMP-3) is a 110-residue mature cystine-knot polypeptide released from a 472-amino-acid pre-proprotein through proprotein convertase cleavage. The functional molecule is a disulfide-linked homodimer whose paired β-sheet fingers present distinct receptor-contact surfaces that preferentially engage activin type II receptors and the ALK7 type I receptor. Constitutively expressed in adult rat trabecular bone, lung alveoli and intestinal submucosa, BMP-3 is secreted as a 25 kDa glycosylated dimer that associates weakly with extracellular matrix heparan sulfate. Antibodies raised against recombinant rat BMP-3 recognize both the mature dimer and the larger precursor with negligible cross-reactivity to BMP-2, BMP-7 or other TGF-β superfamily members, enabling selective immunoblot and immunohistochemical detection in rat tissue extracts and sections.
