Target Information
Rat alpha-hemoglobin-stabilizing protein (AHSP) is a 12 kDa erythroid chaperone whose three-helix bundle clamps free α-globin, preventing its precipitation until β-globin partners arrive. A solvent-exposed 11-aa "QPK" loop (aa 38-48) between helices 2 and 3 presents a β-turn unique among rat globin chaperones, offering an epitope preserved after fixation. Antibodies to this loop detect native AHSP in bone-marrow erythroblasts, circulating reticulocytes, and splenic red pulp, giving crisp cytoplasmic staining and single-band blots without cross-reaction to other chaperones.
