Target Information
Nicotinamide adenine dinucleotide (NAD) is a redox-active coenzyme composed of adenosine monophosphate and nicotinamide mononucleotide linked through a pyrophosphate bridge. The oxidized form, NAD+, accepts a hydride equivalent at the nicotinamide C4 position to generate NADH, creating a reversible redox pair operating at -320 mV. NAD+ also serves as a substrate for ADP-ribosyltransferases and sirtuins, transferring ADP-ribose or acetyl groups to target proteins. Antibodies raised against NAD-protein conjugates recognize the intact dinucleotide with minimal cross-reactivity to NADP, NADH or related nucleotides, allowing direct detection of NAD distribution in fixed cells and tissue homogenates.
