Mouse Wnt-3a is a 39 kDa secreted lipid-modified morphogen whose N-terminal palmitoleoylated serine anchors to Frizzled CRDs, while a C-terminal cystine-knot stabilizes the Wnt-specific β-hairpin required for LRP6 phosphorylation. A solvent-exposed 12-residue "thumb" loop (aa 200-211) houses the signature WGGCX(8)C motif absent from paralogs Wnt-3 and Wnt-3b, offering a selective epitope that survives lipidation and carrier binding.
