Mouse UDP-glucose ceramide glucosyltransferase (UGCG) is a 46 kDa Golgi-resident type-I membrane enzyme whose C-terminal catalytic domain transfers glucose from UDP-Glc to ceramide, initiating glycosphingolipid synthesis. A solvent-exposed 16-aa "KVE" loop (aa 200-215) between transmembrane helix 4 and the UDP-Glc binding pocket folds into a β-hairpin unique among mouse glycosyltransferases, presenting a conformational epitope preserved after membrane isolation and fixation. Affinity-purified antibodies targeting this loop trace native UGCG in hepatic Golgi, neuronal somata, and renal proximal tubules, delivering crisp perinuclear rings and single-band clarity on immunoblots with no cross-reaction to other glucosyltransferases.
