Mouse tumor necrosis factor ligand superfamily member 9 (TNFSF9, 4-1BBL) is a 50 kDa type-II transmembrane glycoprotein whose C-terminal TNF homology domain trimerizes to engage 4-1BB, clustering in lipid rafts without intrinsic catalytic motifs. A solvent-exposed 14-aa "KVE" loop (aa 185-198) between β-strands E and F folds into a β-hairpin unique among mouse TNFSF members, presenting a conformational epitope preserved after trimerization and fixation. Affinity-purified antibodies targeting this loop trace native TNFSF9 in splenic dendritic cells, activated T-cell zones, and bone-marrow stroma, delivering crisp membrane rings and single-band clarity on immunoblots with no cross-reaction to CD40L or other TNFSF ligands.
