Target Information
Mouse tropomyosin 3 (TPM3) is a 284-residue actin-binding protein that forms parallel two-stranded α-helical coiled-coil dimers spanning seven quasi-repeats of 39-42 residues, each presenting a conserved actin-binding consensus. The N-terminus is acetylated and the C-terminus carries a consensus overlap zone that dictates head-to-tail polymerization along F-actin, stabilizing thin filaments and modulating myosin access. Expressed in slow skeletal, cardiac and smooth muscle fibres as well as neurons and fibroblasts, TPM3 localizes to stress fibres, lamellipodia and sarcomeric I-bands. Antibodies raised against the C-terminal peptide or full-length recombinant TPM3 detect native, acetylated and alternatively spliced isoforms with negligible cross-reactivity to TPM1, TPM2 or TPM4, enabling selective immunoblot and immunofluorescence analysis in mouse tissue homogenates and cultured cells.
