Target Information
Mouse sirtuin 2 (SIRT2) is a 389-residue NAD⁺-dependent deacetylase of the sirtuin family, featuring a conserved catalytic Rossmann-fold domain that binds NAD⁺ and a smaller zinc-binding module recognizing acetyl-lysine substrates. Predominantly cytosolic, the enzyme associates with microtubules, centrosomes and oligodendrocyte cytoplasm during mitosis, adopting a cell-cycle-regulated expression peak in G2/M. Its N-terminal extension undergoes phosphorylation, modulating subcellular distribution between cytoplasm and nucleus. Antibodies directed against full-length or peptide epitopes of mouse SIRT2 detect both native and post-translationally modified isoforms with negligible cross-reactivity to SIRT1, SIRT3 or other sirtuins, enabling accurate immunoblot and immunofluorescence analysis in tissue homogenates and cultured cells.
