Target Information
Mouse signal-transducing adaptor molecule 1 (STAM1) is a 540-residue cytosolic scaffold built from an N-terminal VHS domain, a central SH3 module, and a C-terminal ITAM motif that becomes tyrosine-phosphorylated upon cytokine triggering. The protein constitutively localizes to early endosomes via its coiled-coil region, where it assembles with Hrs and eps15 into ESCRT-0-like complexes. Expressed ubiquitously in spleen, thymus and brain, STAM1 cycles between cytosol and clathrin-coated pits. Antibodies raised against the SH3-ITAM linker or full-length recombinant protein detect native, phosphorylated and ubiquitinated isoforms with negligible cross-reactivity to STAM2 or other adaptors, enabling selective immunoblot and immunofluorescence analysis in mouse tissue homogenates and cultured cells.
