Target Information
Mouse signal regulatory protein alpha (SIRPα) is a transmembrane glycoprotein of the immunoglobulin superfamily, a core regulator of immune cell activation and phagocytosis. It features three conserved extracellular Ig-like domains for ligand binding, a single transmembrane segment, and a cytoplasmic tail with immunoreceptor tyrosine-based inhibitory motifs (ITIMs) critical for signaling. It localizes predominantly to the plasma membrane of mouse macrophages, dendritic cells and neurons, with negligible expression in epithelial and erythroid cells. The protein mediates cell-cell interaction via binding to its ligand CD47, with conserved motifs for modulating downstream inhibitory signaling cascades. Mouse SIRPα shares core functional domains with mammalian orthologs, and has distinct regulatory specificity differentiating it from SIRPβ and SIRPγ.
