Target Information

Mouse S-antigen (SAG, arrestin-1) is a 48 kDa soluble protein whose N-terminal immunoglobulin-like domain and C-tail clasp light-activated phosphorhodopsin, locking photoreceptors in an inactive state. A solvent-exposed 14-aa "KVE" loop (aa 203-216) between β-strands 7 and 8 folds into a β-hairpin unique among mouse arrestins, presenting a conformational epitope preserved after rhodopsin binding and fixation. Affinity-purified antibodies targeting this loop trace native SAG in outer-segment layer, pineal gland, and limited forebrain neurons, delivering crisp subretinal puncta and single-band clarity on immunoblots with no cross-reaction to β-arrestin-1 or β-arrestin-2.
