Target Information

Mouse prothrombin fragment 1+2 (F1+2) is the 156-residue activation peptide released upon factor Xa-mediated cleavage of prothrombin at Arg271-Thr272. The fragment comprises the Gla-rich module (residues 1-46) and two kringle domains that together bind calcium and phospholipid, forming a 35 kDa species that circulates transiently before degradation. Constitutively present at low picomolar levels, F1+2 localizes to plasma and thrombin-generating surfaces. Antibodies raised against the Gla-kringle linker or kringle-2 epitope detect native, calcium-bound and proteolyzed forms with negligible cross-reactivity to intact prothrombin or thrombin, enabling selective immunoblot and ELISA quantification in mouse plasma and tissue extracts.
