Mouse protein-tyrosine-phosphatase receptor type H (PTPRH, SAP-1) is a 912-residue transmembrane phosphatase whose extracellular segment carries five fibronectin-type III repeats and a single catalytic domain that dephosphorylates Src-family kinases. A single transmembrane helix anchors the protein to the apical brush border of intestinal epithelium, while a short cytoplasmic tail binds ezrin-radixin-moesin proteins. Constitutively expressed in small intestine and colon, PTPRH localizes to microvilli and tight-junction plaques. Antibodies raised against the ectodomain or catalytic peptide detect native, glycosylated and proteolytically shed forms with negligible cross-reactivity to other receptor PTPs, enabling selective immunoblot and immunohistochemical visualization in mouse intestinal homogenates and sections.
