Target Information
Mouse protein kinase R (PKR) is a 515-residue serine/threonine kinase harboring two N-terminal dsRNA-binding motifs (dsRBM1-2) and a C-terminal kinase domain that autophosphorylates Thr446 and Thr451 upon activation. Constitutively expressed in all tissues, the enzyme localizes to cytoplasm and stress granules, dimerizing upon binding double-stranded RNA or structured cellular RNAs. The latent monomer undergoes conformational rearrangement to expose the active site, which then targets eIF2α and other substrates. Antibodies raised against full-length or phospho-specific epitopes detect native, phosphorylated and cleaved forms with negligible cross-reactivity to other kinases, enabling precise immunoblot and immunofluorescence analysis in mouse tissue homogenates and cultured cells.
