Target Information
Mouse protein kinase D2 (PKD2, PRKD2) is an 878-residue serine/threonine kinase possessing an N-terminal tandem C1A-C1B diacylglycerol-binding module, a central pleckstrin-homology domain for membrane docking, and a C-terminal catalytic core that autophosphorylates Ser707 and Ser711 upon activation. Expressed in spleen, thymus and brain, PKD2 cycles between cytosol and trans-Golgi network, binding phosphatidic acid and diacylglycerol-rich membranes. Antibodies raised against the catalytic domain or phospho-Ser707 detect native, active and cleaved forms with negligible cross-reactivity to PKD1 or PKD3, enabling selective immunoblot and immunofluorescence analysis in mouse tissue homogenates and cultured cells.
