Target Information

Mouse protein kinase D1 (PKD1, PRKD1) is an 875-residue serine/threonine kinase comprising a diacylglycerol-binding C1A-C1B tandem at the N-terminus, a central PH domain for membrane docking, and a C-terminal catalytic core that autophosphorylates Ser744/Ser748 upon activation. Constitutively expressed in brain, heart and salivary glands, PKD1 cycles between cytosol and trans-Golgi network, binding to phosphatidic acid and diacylglycerol-rich membranes. Antibodies raised against the catalytic domain or phospho-Ser744/748 detect native, active and cleaved forms with negligible cross-reactivity to PKD2 or PKD3, enabling selective immunoblot and immunofluorescence analysis in mouse tissue homogenates and cultured cells.
