Mouse proopiomelanocortin (POMC) is a 30-kDa precursor polypeptide predominantly expressed in the arcuate nucleus of the mouse hypothalamus and anterior pituitary gland. It undergoes tissue-specific proteolytic cleavage to produce bioactive peptides-including α-melanocyte-stimulating hormone (α-MSH), adrenocorticotropic hormone (ACTH), and β-endorphin. These peptides regulate key physiological processes: α-MSH suppresses appetite and maintains energy balance via melanocortin receptors, ACTH stimulates adrenal cortisol release, and β-endorphin mediates pain relief and reward. With hypothalamic POMC neurons acting as central metabolic sensors, it integrates nutritional cues to balance metabolism, stress responses, and reproduction. As a critical neuroendocrine precursor, mouse POMC is a target for studying obesity, stress disorders, and endocrine dysfunction.
