Mouse poliovirus receptor-related protein 3 (PRR3, nectin-3) is a 83 kDa single-span Ig superfamily adhesin whose three Ig-like domains bridge homophilic or heterophilic contacts with nectin-1 and -2, stabilizing epithelial and synaptic junctions. A solvent-exposed 16-aa "KVE" loop (aa 142-157) within the second IgC domain adopts a β-turn unique among PRR paralogs, presenting a conformational epitope preserved after formalin fixation. Affinity-purified antibodies targeting this loop trace native PRR3 in Sertoli-cell ectoplasmic junctions, hippocampal dendritic spines, and renal tubule epithelia, delivering crisp membrane doublets and single-band specificity on immunoblots with no cross-reaction to PRR1 or PRR2.
