Target Information

Mouse paxillin is a 559-residue cytoskeletal adaptor built from an N-terminal polyproline stretch, five tandem LD motifs that dock integrin-linked kinase and focal adhesion kinase, followed by four cysteine-rich LIM domains that anchor to actin and vinculin. Phosphorylatable tyrosines at residues 31 and 118 create tandem SH2-binding sites upon Src-family kinase engagement. Constitutively expressed in fibroblasts, epithelia and smooth muscle, paxillin localizes to focal adhesions, costameres and podosomes. Polyclonal and monoclonal antibodies raised against full-length or peptide epitopes detect both native and phosphorylated isoforms with negligible cross-reactivity to Hic-5 or leupaxin, enabling precise immunoblot and immunofluorescence analysis in mouse tissue extracts and cultured cells.
