Target Information
Mouse palmitoyl protein thioesterase 1 (PPT1) is a 306-residue soluble glycoprotein that removes fatty acyl chains from S-acylated cysteines. Synthesized with a 26-aa signal peptide, the mature 27 kDa enzyme folds into an α/β hydrolase domain with a catalytic triad (Ser115, Asp233, His289) and six N-linked glycans that stabilize a compact dimer. Constitutively expressed in neurons, glia and visceral epithelia, PPT1 localizes to lysosomal lumen via mannose-6-phosphate tagging. Antibodies raised against recombinant full-length mouse PPT1 detect native, glycosylated and proteolytically processed forms with negligible cross-reactivity to PPT2 or other thioesterases, enabling precise immunoblot and immunohistochemical visualization in mouse tissue homogenates and sections.
