Mouse nuclear factor, erythroid-derived 2-like protein 2 (Nrf2) is a 597-residue transcription factor harboring six Neh domains: an N-terminal BTB-Kelch binding region, two transactivation segments rich in hydrophobic and acidic residues, a central CNC-bZIP DNA-binding module, and a C-terminal Kelch-repression site. Under basal conditions its DLG and ETGE motifs tether to Keap1, targeting the protein for ubiquitination; oxidative modification releases Nrf2, permitting nuclear accumulation. Constitutively expressed in liver, kidney and airway epithelium, the factor recognizes antioxidant response elements bearing the consensus TGAnnnnGC sequence. Antibodies raised against full-length or Neh1-specific peptides detect native, phosphorylated and cleaved isoforms with negligible cross-reactivity to Nrf1 or Nrf3, enabling precise immunoblot and immunohistochemical analysis in mouse tissue homogenates and sections.
