Target Information
Mouse neutrophil cytosolic factor 4 (NCF4, p40-phox) is a 39 kDa PX-SH3 adaptor whose N-terminal phosphoinositide-binding pocket and C-terminal SH3 domain bridge p67-phox to membrane gp91-phox, assembling the NADPH oxidase that generates microbicidal superoxide. A solvent-exposed 16-aa "KVE" loop (aa 190-205) between the PX and tandem SH3 modules folds into a β-hairpin unique among mouse oxidase subunits, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native NCF4 in bone-marrow neutrophils, splenic macrophages, and stimulated peritoneal exudates, delivering crisp cytoplasmic puncta and single-band clarity on immunoblots with no cross-reaction to NCF1 or NCF2.
