Mouse neuronal apoptosis inhibitory protein (NAIP) is a 140 kDa cytosolic NLR whose three N-terminal baculoviral repeats grip activated caspases, while a C-terminal nucleotide-binding domain assembles inflammasome platforms. A solvent-exposed 18-aa "KVE" loop (aa 420-437) between BIR2 and BIR3 folds into a β-hairpin unique among mouse NLR proteins, presenting a conformational epitope preserved after aldehyde fixation. Affinity-purified antibodies targeting this loop trace native NAIP in spinal motor neurons, hippocampal pyramidal layers, and macrophage cytoplasm, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to other IAP family members.
