Mouse mitogen-activated protein kinase 7 (MAPK7, ERK5) is a 98 kDa dual-specificity kinase whose N-terminal kinase domain phosphorylates Ser/Thr targets, while a unique C-terminal tail binds transcription factors, driving growth and stress responses. A solvent-exposed 18-aa "KVE" loop (aa 475-492) between the activation segment and C-tail folds into a β-hairpin unique among mouse MAPK isoforms, presenting a conformational epitope preserved after phosphorylation and fixation. Affinity-purified antibodies targeting this loop trace native MAPK7 in vascular endothelium, cardiac myocytes, and neuronal nuclei, delivering crisp cytoplasmic/nuclear staining and single-band clarity on immunoblots with no cross-reaction to ERK1 or ERK2.
