Mouse membrane-spanning 4-domains subfamily A member 1 (MS4A1), also termed CD20, is a 297-residue tetraspan phosphoprotein that threads through the plasma membrane four times, creating two extracellular loops of 42 and 72 residues and cytoplasmic N- and C-termini. The larger loop harbors an evolutionarily conserved N-linked glycosylation site and a disulfide bond that stabilize a compact α-helical bundle. Expressed exclusively on the surface of developing and mature B lymphocytes, MS4A1 localizes to lipid rafts and microvilli. Antibodies raised against the larger extracellular loop detect native MS4A1 with negligible cross-reactivity to other MS4A family members, enabling precise immunoblot, immunohistochemistry and flow-cytometric detection in mouse lymphoid tissues and cultured B cells.
