Mouse membrane protein, palmitoylated 2 (MPP2, also called palmitoylated membrane protein 2) is a 539-residue member of the MAGUK family, built from two N-terminal L27 domains, a central PDZ module, an SH3 motif, and a C-terminal guanylate kinase-like region that lacks catalytic activity. Dual palmitoylation at Cys 5/6 anchors the polypeptide to the inner leaflet of postsynaptic and epithelial plasma membranes, where it assembles scaffold complexes via PDZ-mediated interactions. Constitutively expressed in brain, retina and kidney, MPP2 localizes to synaptic densities and tight-junction plaques. Antibodies raised against the PDZ-SH3 linker or C-terminal peptide detect native, palmitoylated and phosphorylated isoforms with negligible cross-reactivity to other MAGUKs, enabling selective immunoblot and immunofluorescence in mouse tissue homogenates and cultured cells.
