Target Information
Mouse MAPK-activated protein kinase 2 (MK2) is a 400-residue serine/threonine kinase containing an N-terminal catalytic domain and a C-terminal autoinhibitory helix that unfolds upon p38 MAPK binding. Phosphorylation of Thr334 within the activation loop triggers catalytic activity toward Hsp27 and other substrates. Constitutively expressed in brain, lung and spleen, MK2 localizes to nucleus and cytoplasm, shuttling between compartments in a stress-dependent manner. Antibodies raised against full-length or phospho-Thr334 peptide detect native, phosphorylated and truncated isoforms with negligible cross-reactivity to MK3 or other kinases, enabling selective immunoblot and immunofluorescence analysis in mouse tissue homogenates and cultured cells.
