Target Information
Mouse Kelch-like ECH-associated protein 1 (Keap1) is a 624-residue cytosolic repressor built from an N-terminal BTB domain that mediates Cul3 ubiquitin ligase binding, a central intervening region rich in reactive cysteines, and a C-terminal Kelch repeat propeller providing two binding pockets for the DLG and ETGE motifs of Nrf2. The dimeric scaffold adopts a cherry-bob conformation, positioning substrate and ubiquitin machinery 20 Å apart. Constitutively expressed in liver, kidney and airway epithelium, Keap1 localizes to perinuclear actin and focal adhesions. Antibodies raised against full-length or Kelch-specific peptides detect native, phosphorylated and oxidized forms with negligible cross-reactivity to other BTB-Kelch proteins, enabling selective immunoblot and immunofluorescence detection in mouse tissue and cultured cells.
