Mouse inhibitory subunit of NF-κB alpha (IκBα) is a 37-kDa cytoplasmic protein ubiquitously expressed in mouse tissues and immune cells. It functions as a key regulator of NF-κB signaling, sequestering NF-κB dimers in the cytoplasm to inhibit their nuclear translocation. Upon pro-inflammatory stimuli (e.g., TNF-α, LPS) or pathogen recognition, IκBα undergoes phosphorylation and proteasomal degradation, releasing NF-κB to activate transcription of pro-inflammatory cytokines, chemokines, and survival genes. Critical for balancing murine immune activation, inflammation, and tissue homeostasis, its dysregulation links to autoimmune diseases, cancer, and infectious pathologies. As a central modulator of NF-κB signaling, mouse IκBα is a critical target for studying immunology, inflammatory disorders, and signal transduction.
