Mouse IKK-interacting protein (IKIP) is a 28 kDa cytosolic scaffold that clamps the IKKα/β catalytic core to the regulatory NEMO subunit, aligning the kinase active sites with IκB substrates. A solvent-exposed 17-residue "KAL" loop (aa 92-108) between coil-2 and coil-3 folds into a β-hairpin unique among IKK-associated factors, offering a fixation-stable epitope preserved under native and denaturing conditions. Affinity-purified antibodies targeting this loop trace endogenous IKIP in splenic B cells, macrophage cytoplasm, and fibroblast stress fibers, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to IKKα, IKKβ, or NEMO.
