Mouse Hermansky Pudlak Syndrome Protein 4 (HPS4) is a cytoplasmic subunit of the biogenesis of lysosome-related organelles complex-3 (BLOC-3), a key mediator of lysosome-related organelle (LRO) biogenesis and trafficking. It features conserved N-terminal and C-terminal interaction domains, a central structural region, and motifs critical for complex assembly and stability. It localizes to the cytoplasm and late endosomes of mouse melanocytes, platelets and lung epithelial cells, with negligible levels in muscle and neuronal tissues. The protein mediates LRO cargo sorting and organelle maturation, with conserved binding sites for HPS1 and other BLOC-3 components. Mouse HPS4 shares core functional domains with mammalian orthologs, and has distinct tissue specificity differentiating it from other HPS family proteins.
