Mouse H2A histone family member Z (H2A.Z) is a 14 kDa replacement histone whose extended αC-helix and three unique surface residues dock into nucleosomes, destabilizing DNA wrapping and marking active or poised chromatin. A solvent-exposed 12-aa "KVE" loop (aa 85-96) between αC and α3 folds into a β-hairpin unique among mouse H2A variants, presenting a conformational epitope preserved after nucleosome assembly and fixation. Affinity-purified antibodies targeting this loop trace native H2A.Z in euchromatic foci, transcriptional start sites, and pericentric heterochromatin, delivering crisp nuclear puncta and single-band clarity on immunoblots with no cross-reaction to canonical H2A or H2A.X.
