Mouse guanidinoacetate-N-methyltransferase (GAMT) is a cytosolic methyltransferase with a conserved S-adenosyl-L-methionine (SAM)-binding domain, a core mediator of creatine biosynthesis. It features an N-terminal catalytic region critical for methyl group transfer, a central substrate-binding motif for guanidinoacetate recognition, and a C-terminal domain for structural stability. It localizes predominantly to the cytoplasm of mouse hepatocytes and renal tubular cells, with negligible expression in muscle or neuronal tissues. The protein mediates the key methylation step in creatine production, with conserved structural elements differentiating it from other SAM-dependent methyltransferases.
