Target Information
Mouse growth differentiation factor 10 (GDF-10, BMP-3b) is a 52 kDa disulfide-linked homodimer whose C-terminal TGF-β knot binds BMPR-II and ALK3/6 to restrain osteoblast maturation and promote cartilage homeostasis. A solvent-exposed 16-residue "KVE" loop (aa 102-117) between pre-helix 1 and β2 adopts a β-hairpin unique among mouse GDF/BMP isoforms, presenting a conformational epitope preserved in both pro- and mature forms. Affinity-purified antibodies targeting this loop trace native GDF-10 in calvarial osteocytes, periodontal ligament, and neonatal cartilage, delivering crisp pericellular staining and single-band clarity on immunoblots with no cross-reaction to BMP-2 or GDF-5.
