Mouse growth arrest and DNA-damage-inducible protein gamma (GADD45G) is an 18 kDa intrinsically disordered acid blob that clamps onto the active site of PCNA, transiently blocking replicative polymerases while recruiting repair endonucleases. Its N-terminal 33 residues fold into a U-shaped β-hairpin that docks with MTK1 and p38 MAPK, transmitting stress cues without phosphorylation. A solvent-exposed DE-loop (aa 51-65) houses a conformation-specific epitope absent in paralogs GADD45A and GADD45B, enabling polyclonal antibodies to trace nuclear foci, cytoplasmic stress granules, and serum-derived exosomes with single-band specificity on immunoblots.
