Mouse glycophorin A (GYPA) is a 15 kDa single-pass sialoglycoprotein whose extracellular Ser/Thr-rich segment carries 10 O-linked sugar chains that generate a negatively charged glycocalyx coat. A solvent-exposed 12-residue "STD" motif (aa 58-69) within the hyper-glycosylated region folds into a strain-unique β-turn, creating an epitope preserved after neuraminidase treatment and formalin fixation. Affinity-purified antibodies targeting this motif trace native GYPA in erythrocyte ghosts, bone-marrow erythroblasts, and spleen sinusoids, delivering crisp membrane rings and single-band clarity on immunoblots with no cross-reaction to glycophorin C or band 3.
