Target Information
Mouse glucokinase regulatory protein (GKRP) is a 626-residue nucleocytoplasmic shuttling protein that binds hepatic glucokinase (GCK) in the nucleus under low-glucose conditions. Its N-terminal 18-residue tail contains a bipartite nuclear-localization signal, while a central 300-residue GCK-binding domain forms a hydrophobic pocket that stabilizes the inactive enzyme conformation. Expressed almost exclusively in liver and pancreatic islets, GKRP localizes to hepatocyte nuclei and translocates to the cytoplasm at high glucose concentrations. Antibodies raised against full-length or N-terminal peptide detect native, phosphorylated and ubiquitinated isoforms with negligible cross-reactivity to GCK or other nuclear receptors, enabling selective immunoblot and immunofluorescence analysis in mouse liver homogenates and cultured hepatocytes.
