Mouse free fatty acid receptor 2 (FFAR2, GPR43) is a 35-kDa G protein-coupled receptor predominantly expressed in murine intestinal epithelium, immune cells (neutrophils, macrophages), and adipose tissue. It binds short-chain fatty acids (SCFAs, e.g., acetate, propionate) to activate Gαi/Gαq signaling, regulating gut barrier function, immune modulation, and metabolic homeostasis. Critical for murine gut microbiota-host crosstalk, it modulates inflammation, glucose metabolism, and appetite regulation. Dysregulated expression associates with inflammatory bowel disease, obesity, and metabolic syndrome in mouse models, as impaired SCFA signaling disrupts gut-immune-metabolic crosstalk. As a key mediator of microbial metabolite signaling, mouse FFAR2 is a critical target for studying gut-brain-axis biology and developing therapeutics for metabolic and inflammatory disorders.
