Mouse FK506-binding-protein-like (FKBPL) is a 42 kDa secreted immunophilin whose N-terminal amphipathic helix docks onto Hsp90 chaperone complexes, while the C-terminal super-helix clamps cell-surface CD44 to restrain actin-driven motility. A solvent-exposed LKXE motif, unique among FKBP paralogs, provides a high-affinity epitope that survives formalin fixation and antigen retrieval. Polyclonal antibodies raised against this signature sequence trace FKBPL from endoplasmic-reticulum exit sites through Golgi vesicles to extracellular clusters along basement membranes, distinguishing native protein from smaller cytosolic splice variants without cross-reacting with classical FKBP12.
