Mouse ficolin-2 is a 35 kDa secreted pattern-recognition protein whose collagen-like stalk and C-terminal fibrinogen-related domain bind GlcNAc and acetylated sugars on microbial surfaces, triggering lectin-complement pathway activation. A solvent-exposed 16-aa "KVE" loop (aa 210-225) between the collagen and neck regions folds into a β-hairpin unique among murine ficolins, presenting a conformational epitope preserved after glycosylation and fixation. Affinity-purified antibodies targeting this loop trace native ficolin-2 in serum, hepatic sinusoids, and bronchoalveolar lavage, delivering crisp extracellular bands and single-band clarity on immunoblots with no cross-reaction to ficolin-1 or ficolin-3.
