Mouse EDA is a type II transmembrane ligand protein containing a conserved tumor necrosis factor homology domain at its extracellular terminus. It maintains stable trimeric conformation and intact receptor-interacting motifs under physiological conditions, with soluble cleaved variants circulating in extracellular fluid. Its dual membrane-tethered and secreted isoforms, plus unique trimeric structural fold, set it apart from cytoplasmic catalytic enzymes, organelle transporters and intranuclear transcription factors that carry simpler domain arrangements and static subcellular localization profiles.
