Mouse dickkopf-related protein 3 (DKK3) is a 38 kDa secreted glycoprotein whose N-terminal cysteine-rich region and C-terminal coiled-coil domain adopt a compact fold that sterically hinders Wnt-receptor engagement without direct binding. A solvent-exposed 16-aa "KVE" loop (aa 165-180) between the second and third disulfide bridges folds into a β-hairpin unique among murine Dkk family members, presenting a conformational epitope preserved after secretion and fixation. Affinity-purified antibodies targeting this loop trace native DKK3 in retinal pigment epithelium, cardiac fibroblasts, and pulmonary smooth-muscle layers, delivering crisp extracellular deposition and single-band clarity on immunoblots with no cross-reaction to DKK1 or DKK2.
