Target Information
Mouse diacylglycerol kinase zeta (DGKζ) is a cytoplasmic serine/threonine kinase of the DGK family, a core regulator of lipid signaling and intracellular second messenger homeostasis. It features a conserved N-terminal catalytic domain with ATP-binding motif, tandem cysteine-rich C1 domains for diacylglycerol (DAG) binding, and a C-terminal PDZ-binding motif critical for protein-protein interaction. It localizes to the cytoplasm and plasma membrane of mouse immune, neuronal and epithelial cells, with negligible levels in terminally differentiated tissues. The protein mediates DAG phosphorylation to phosphatidic acid (PA), with conserved motifs for modulating PKC and Ras signaling pathways. Mouse DGKζ shares core functional domains with mammalian orthologs, and has distinct tissue specificity differentiating it from DGKα and DGKε.
