Mouse Cytochrome P450 24A1 (CYP24A1) is a mitochondrial inner-membrane enzyme that terminates vitamin D signaling by hydroxylating 1,25-dihydroxyvitamin D₃ and 25-hydroxyvitamin D₃ at positions C-24 and C-23, generating calcitroic acid and 24,25-dihydroxyvitamin D₃ for biliary excretion. Constitutively expressed in kidney, intestine, and bone, its activity is rapidly up-regulated by calcitriol-bound VDR, forming a negative-feedback loop that prevents vitamin D intoxication and maintains calcium-phosphate homeostasis. Antibodies to CYP24A1 enable precise localization of vitamin D catabolic activity in mouse kidney sections, primary proximal-tubule cells, and metabolic studies without cross-reactivity to other P450 isoforms .
