Target Information
Mouse complement C3 convertase is a 165 kDa C4bC2a heterodimer whose C2a serine-protease active site cleaves C3 into C3a and C3b, amplifying complement cascade on pathogen and host surfaces. A solvent-exposed 16-aa "KVE" loop (aa 285-300) within the C2a catalytic cleft folds into a β-hairpin unique among mouse complement enzymes, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native C3 convertase in hepatic sinusoids, splenic follicles, and glomerular capillaries, delivering crisp surface deposition and single-band clarity on immunoblots with no cross-reaction to C5 convertase or factor B.
