Target Information
Mouse coagulation factor XIII A1 polypeptide (F13A1) is a 730-residue cytosolic transglutaminase comprising an N-terminal β-sandwich, a catalytic core with active-site Cys314, and two C-terminal β-barrel domains that engulf Ca²⁺ and protein substrates. Upon thrombin cleavage of an N-terminal 37-residue activation peptide, the 83 kDa monomer assembles into a Ca²⁺-dependent homodimer that cross-links γ-glutamyl-ε-lysyl isopeptide bonds in fibrin and extracellular matrix. Constitutively expressed in platelets, monocytes and osteoblasts, F13A1 localizes to cytoplasm and cell surface after activation. Antibodies raised against full-length or thrombin-cleaved forms detect native, activated and cross-linked species with negligible cross-reactivity to tissue transglutaminase, enabling selective immunoblot and immunohistochemical detection in mouse plasma and tissue homogenates.
