Target Information
Mouse cluster of differentiation 72 (CD72) is a 359-residue type II transmembrane glycoprotein whose extracellular C-terminal region carries two C-type lectin-like domains that bind CD100 (Semaphorin-4D). A single transmembrane helix anchors the protein to B-cell and dendritic-cell plasma membranes, while a 61-aa N-terminal cytoplasmic tail contains two ITIM motifs that recruit SHP-1 and SHP-2 upon phosphorylation. Constitutively expressed in spleen, lymph node and bone-marrow, CD72 localizes to lipid rafts and the immunological synapse. Antibodies raised against the lectin-like or cytoplasmic peptide detect native, glycosylated and phosphorylated isoforms with negligible cross-reactivity to other lectin receptors, enabling selective immunoblot and immunofluorescence analysis in mouse lymphoid tissue homogenates and cultured immune cells.
