Target Information
Mouse cluster of differentiation 28 (CD28) is a 243-residue type I transmembrane glycoprotein whose extracellular V-like Ig domain (residues 20-120) binds the MYPPPY motif in B7-1 (CD80) and B7-2 (CD86). A single transmembrane helix anchors the protein to T-cell surfaces, while a 41-aa cytoplasmic tail carries two YXN motifs that recruit PI3K and GRB2 after phosphorylation. Constitutively expressed on CD4⁺ and CD8⁺ T cells, CD28 localizes to the immunological synapse and microvilli. Antibodies raised against the IgV or cytoplasmic peptide detect native, glycosylated and phosphorylated isoforms with negligible cross-reactivity to CTLA-4 or other CD28 family members, enabling selective immunoblot and immunofluorescence analysis in mouse lymphoid tissue homogenates and cultured T cells.
